Mueller was named by Deputy Attorney General Rod Rosenstein eight days after Comey's dismissal to lead the Russian Federation investigation as a special counsel, a position created to conduct investigations when a normal Justice Department probe would present a conflict of interest or in other extraordinary circumstances.
U.S. Sen. Dianne Feinstein, a Democrat from California, said she is becoming "increasingly concerned" that Trump will terminate both Mueller and Rosenstein.
Trump and his supporters cast Comey's testimony that he had told the president he was not personally under investigation as vindication.
Trump appeared to confirm he was being investigated for obstruction in a tweet on Friday that read: "I am being investigated for firing the FBI Director by the man who told me to fire the FBI Director!"
But Robert Mueller has been appointed special counsel to investigate Russian involvement in the 2016 presidential election and possible collusion with the Trump campaign.
The president has denied trying to influence the investigation but has acknowledged in an interview that Russian Federation was on his mind when he dismissed Comey.
Regardless, Trump probably wants to fire Mueller now more than he ever has. "Witch Hunt", Trump wrote on Twitter, referring to his May 9 dismissal of James Comey.
Trump's digital volley appeared to be trained at Deputy Attorney General Rod Rosenstein, who recommended the firing last month of James Comey - an nearly unprecedented move that could yet imperil Trump's presidency.

Sources told ABC News that Rosenstein raised the possibility of his own recusal during a meeting with Associate Attorney General Rachel Brand, who's next in line at the Department of Justice after Rosenstein. The aide spoke on condition of anonymity, because the meeting was private.
"Crooked Hillary destroyed phones with hammer, "bleached" emails, and had husband meet with Attorney General days before she was cleared - and they talk about obstruction?" he had said. Trump initially used that memo along with a recommendation from Sessions to justify firing Comey.
Meanwhile, The New York Times reported on June 16 that the general counsel's office of Trump's transition team has ordered staff to preserve all materials relating to Russian Federation or Ukraine, including any possible travel documents. Investigators are also considering the conversations Comey and the president had leading up to that point.
The Washington Post has said Mueller was seeking interviews with three Trump administration officials not involved in Trump's campaign: Dan Coats, the director of national intelligence; Michael Rogers, the head of the National Security Agency; and Richard Ledgett, the former NSA deputy director, the AP said.
Trump's personal lawyer, Marc Kasowitz, said in a statement that the president felt he was fully vindicated by Comey's testimony, pointing to Comey saying the president wasn't directly under investigation.
The president, while boarding Marine One on the White House South Lawn Friday morning en route to Miami ignored a reporter's question as to whether he still has faith in Rosenstein.
In a related matter, The Washington Post reported that Mueller is also looking into the finances and business dealings of presidential adviser Jared Kushner, citing officials familiar with the matter.
Sessions had already recused himself from the investigation, testifying earlier this week that he did so because he had worked on the Trump campaign.
