U.S. Secretary of State Mike Pompeo has alleged the ship would head to Tartous to offload its 2.1 million barrels of Iranian crude oil, worth some $130 million.
The actions of the Adrian Darya follow a pattern of other Iranian ships turning off their trackers once they reach near Cyprus in the Mediterranean Sea, said Ranjith Raja, a lead analyst at the data firm Refinitiv.
Earlier on Tuesday, Iran's President Hassan Rouhani ruled out holding any direct talks with the United States, and warned that Tehran would stop complying with other elements of the 2015 accord if the talks with European nations yield no results by Thursday.
Rouhani told lawmakers that Iran wouldn't negotiate directly with the USA unless it returned to the deal.
Previously, the ship-tracking website MarineTraffic.com showed the Adrian Darya off Lebanon on a course toward Tartous, Syria.
A second Iranian official said: "Although the European Union and particularly France have goodwill, they should convince the U.S.to cooperate with them". "Foreigners usually have their own take but our policy guidelines are set by the Supreme Leader".
The vessel, formerly named Grace 1, was detained by British Royal Marine commandos off Gibraltar on July 4 as it was suspected to be en route to Syria in violation of European Union sanctions.
He further described Iran's relations with Russian Federation to be at an acceptable level, adding that an Iranian delegation have gone to France to test level of European Union countries' commitments to the JCPOA.

The United States imposed sanctions on Iran's civilian space agency and two research organizations on Tuesday, saying they were being used to advance Tehran's ballistic missile program.
Iran has warned it will take additional steps away from the accord on Friday if it doesn't get help from Europe to sell its oil overseas, calling it their "third step" away from the deal.
Tehran has threatened to reduce its nuclear commitments further by September 6, which would include enriching uranium to 20%, and restarting the centrifuges that purify uranium to weapons' grade.
A French diplomatic source said it was important, after recent discussions between Paris and Tehran, to establish that Rouhani was "still ready to negotiate". That would allow Iran to obtain foreign hard currency.
"That all assuming that President Trump allows waivers on some points" of the new USA sanctions on Iran, he added.
"Unfortunately after the United States betrayal. the Europeans haven't acted on their commitments or couldn't.in some cases they could have acted but did not", he said.
"However, if they fail to take any significant measure, we will definitely take the third step in the coming days", the president underlined.
As part of their efforts to keep the nuclear deal alive, France, Germany and Britain have set up a mechanism called INSTEX that would allow continued trade with Iran despite the U.S. sanctions.
